Product Specific Information

The OKT3 monoclonal antibody recognizes an epitope on the epsilon-subunit within the human CD3 complex. It has potent immunosuppressive properties and has been used effectively in renal, heart, and liver allograft rejection treatments. CD3 is expressed by thymocytes and mature T cells and is essential for TCR complex assembly and activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells
Recommended use: 5 µL (0.2 µg) per test (100 µL total volume, 10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample prior to combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample.

Target Information

The CD3 complex transduces antigen-recognition signals into the cytoplasm of T cells and regulates TCR surface expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits, encoded by genes on chromosome 11. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
