Target Information
Calcineurin binding protein (Cabin-1) and its rat homolog, Cain, are nuclear phosphoproteins that regulate the serine/threonine phosphatase activity of calcineurin, influencing T cell signaling and apoptosis. Calcineurin is essential for transcriptional activation of cytokines and transcription factors such as NFAT, NFκB, and AP-1 involved in T cell receptor (TCR)-mediated signaling.
TCR activation triggers PKC-mediated hyperphosphorylation of Cabin-1, promoting its high-affinity binding to calcineurin. This complex inhibits calcineurin activity and attenuates TCR-mediated signaling. Cabin-1 also interacts with MEF-2 transcription factors, leading to inhibition of MEF-2-mediated gene transcription and apoptosis.
