Target Information
Von Hippel-Lindau syndrome (VHL) is a dominantly inherited familial cancer syndrome that predisposes individuals to various malignant and benign tumors. Germline mutations of the VHL gene are responsible for the familial inheritance of VHL syndrome. The encoded protein is part of a complex including elongin B, elongin C, and cullin-2, possessing ubiquitin ligase E3 activity. It mediates ubiquitination and degradation of hypoxia-inducible factor (HIF), a transcription factor central to oxygen-regulated gene expression. RNA polymerase II subunit POLR2G/RPB7 is also reported as a target. Alternatively spliced transcript variants encoding distinct isoforms have been observed.
