Target Information
DNA methylation, the addition of methyl groups to cytosine bases in CpG dinucleotides, is essential for proper development and gene regulation. The demethylation enzyme associated with the methyl-CpG-binding domain (MBD) can revert methylated cytosine to cytosine by cleaving the methyl group as methanol.
MeCP2 and MBD1 repress transcription by binding to methylated DNA, while MBD2 and MBD4 (also known as MED1) colocalize with heavily methylated satellite DNA and mediate methylation signaling. MBD3 does not bind methylated DNA.
MBD1–4 are expressed in somatic tissues, but MBD1 and MBD2 expression is reduced or absent in embryonic stem cells. MBD4 shares homology with bacterial base excision repair DNA N-glycosylases/lyases and can be mutated in microsatellite unstable tumors.
