Product Specific Information
This HLNC4 monoclonal antibody reacts with human poly (ADP-ribose) polymerase (PARP1), a 116 kDa nuclear enzyme involved in DNA repair. During apoptosis, caspases -3, -6, and -7 cleave PARP1 after Asp214, generating 85 kDa and 25 kDa fragments and inactivating PARP1.
The HLNC4 antibody specifically recognizes the 85 kDa fragment produced after cleavage and does not bind the full-length 116 kDa protein.
Immunogen peptide: NH₂-GVDEVAKKKSKKEKDC-COOH.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry using staurosporine-stimulated Jurkat cells with the Foxp3/Transcription Factor Buffer Set (Product # 00-5523-00).
Recommended protocol: BestProtocols® – Intracellular Staining (Protocol B).
- Recommended use: 5 µL (0.015 µg) per test (100 µL final volume)
- Cell number: 10⁵–10⁸ cells/test (empirically determined)
- Excitation: 488 nm; Emission: 519 nm; Laser: Blue Laser

Target Information
Poly ADP-Ribose Polymerase (PARP) catalyzes ADP-ribose transfer from NAD⁺ to nuclear protein acceptors. Cleavage into an 85 kDa fragment is an early apoptosis marker mediated by caspase CPP32, occurring between Asp216 and Gly217, a conserved site across species.
