Product Specific Information

The YN1/1.7.4 monoclonal antibody reacts with mouse CD54 (ICAM-1), a 90–110 kDa transmembrane glycoprotein expressed by monocytes, lymphocytes, dendritic cells, and endothelial cells. Expression is upregulated on activated lymphocytes, playing a key role in inflammatory responses through interactions with CD11a and CD11b.

• Applications Tested: Mouse splenocytes, ≤ 0.4 µg/test (in 100 µL final volume, 10⁵–10⁸ cells/test)

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample (regardless of the number of NovaFluors) to antibody cocktails before combining with cells.
• For single-color controls: 5 µL of CellBlox per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures optimized for minimal spillover and spread.

Internal testing shows NovaFluor Yellow 700 non-specifically stains B cells in SJL mice.
No non-specific staining observed in BALB/c or C57BL/6 mice.
Other strains not tested.

Target Information

ICAM-1 (CD54) is an 85–110 kDa type I membrane glycoprotein comprising five immunoglobulin-like extracellular domains, a transmembrane region, and a cytoplasmic tail. It binds integrins CD11a/CD18 (LFA-1) and CD11b/CD18 (Mac-1) and serves as a receptor for rhinovirus.
Expressed on activated endothelial cells, epithelial cells, fibroblasts, chondrocytes, and various immune cells. Soluble ICAM-1 levels increase in inflammatory conditions.
