Target Information

Dectin-2 is a C-type lectin expressed by dendritic cells, macrophages, and inflammatory monocytes. It is a type II membrane receptor with a short cytoplasmic tail and a C-terminal C-type lectin domain. Dectin-2 recognizes high mannose structures and mediates cytokine production from dendritic cells and other antigen-presenting cells.

Although Dectin-2 lacks obvious signaling motifs in its cytoplasmic tail, it utilizes FcRγ for internalization and activation of the NF-κB pathway to induce cytokine production. Despite sharing only 20–25% homology with Dectin-1, it functions similarly as a pattern recognition receptor (PRR) for fungal infections, activating both innate (Syk-CARD9 pathway) and adaptive immune responses through dendritic cell activation.
Thus, Dectin-2 serves as a connector molecule between innate and adaptive immunity at the dendritic cell level.
