Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon activation, p53 induces expression of genes such as p21waf1, leading to G1 or G2/M arrest.
The structure of p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at sites such as Ser15 and Ser392 regulates activation and stabilization, with Ser392 phosphorylation associated with tumor formation. p53 is expressed at low levels in normal cells but is upregulated in transformed cells, contributing to malignancy. Mutations in the TP53 gene cause loss of tumor suppressor activity and are common in many cancers, including hereditary syndromes like Li-Fraumeni.
