Description:
The eBio124-1D1 monoclonal antibody reacts with human CD7, also known as gp40 and Leu9. CD7 is a 40 kDa receptor and a member of the immunoglobulin gene superfamily. The N-terminal amino acid sequence (aa1–107) is highly homologous to Ig kappa light chain sequence, while the carboxyl-terminal region is proline-rich and forms a stalk projecting the Ig domain.

CD7 is expressed on most immature and mature T lymphocytes and T cell leukemias, as well as on natural killer cells, a small subset of normal B cells, and malignant B cells. Cross-linking of surface CD7 modulates T cell and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 directly associates with phosphoinositol 3′-kinase, leading to D-3 phosphoinositide production and tyrosine phosphorylation upon ligation.

A clonogenic subset of human CD34(+) CD38(-) cord blood cells expressing CD45RA, HLA-DR, and high levels of CD7 has been identified as lymphoid progenitors capable of generating B cells, NK cells, and dendritic cells, but not myeloid or erythroid cells. CD7(+) phenotype distinguishes primitive human lymphoid progenitors from pluripotent stem cells.

Additionally, CD7 may cooperate with CD28 in Treg function; mice lacking both CD28 and CD7 exhibit reduced Treg populations and suppressive activity.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage ≤1 µg per test (100 µL final volume). Cell numbers may range from 10⁵ to 10⁸ cells/test. Optimal titration is recommended.

Filtration:
0.2 µm post-manufacturing filtered.
