ROCK1은 세린/트레오닌 키나아제로, 세포 분화 및 이동을 조절합니다. Rho/ROCK 경로는 다양한 인간 암의 진행에 관여하며, ROCK1은 Caspase-3의 새로운 기질로 알려져 있습니다.
ROCK은 소형 GTPase Rho의 효과 단백질 중 하나로, myosin light chain (MLC)의 직접 인산화 및 MLC phosphatase의 인산화를 통해 세포 수축성을 증가시킵니다. 이는 세포사멸(apoptosis) 과정에서 막 융기(membrane blebbing)에 필요한 MLC 인산화의 지속적 증가를 유도합니다.
ROCK-1은 N-말단의 kinase 도메인과 C-말단의 억제성 cysteine/histidine-rich 도메인으로 구성되며, 이들은 Rho-binding 도메인을 포함하는 가변 영역으로 연결됩니다. 세포사멸 중 Caspase-3는 ROCK-I을 절단하여 억제 도메인을 제거하고, 지속적으로 활성화된 kinase 활성을 유도합니다. 이 과정은 MLC의 인산화 및 세포막 융기를 유발하며, 이는 세포사멸의 초기 단계에서 중요한 역할을 합니다.
