Target Information

ALAS1 is a nuclear-encoded mitochondrial matrix enzyme that catalyzes the condensation of glycine with succinyl-CoA to form δ-aminolevulinate, CO₂, and CoA.
It regulates the first and rate-limiting step of the heme biosynthetic pathway and serves as a pyridoxal-5′-phosphate-dependent housekeeping enzyme.
ALAS1 is ubiquitously expressed, providing heme for cytochromes and other hemoproteins. In the liver, ALAS1 is negatively regulated by heme.
Transcription of the ALAS1 gene is stimulated by cAMP and respiratory uncoupling, while phorbol esters and insulin repress its expression.
ALAS1 may play a role in the reciprocal regulation of heme biosynthesis and the circadian clock, suggesting a potential target for circadian disorder treatment.
Downregulation of ALAS1 in acute liver failure leads to altered heme metabolism and impaired liver function.
