Target Information
Neuronal PAS domain protein 3 (NPAS3) is a brain-enriched basic helix-loop-helix PAS domain transcription factor broadly expressed in the developing neuroepithelium. It has been linked to genetic translocation in families affected by schizophrenia. NPAS3 regulates FGF signaling in the dentate gyrus by controlling FGF receptor subtype 1 expression, influencing neurogenesis. NPAS3 knockout mice show growth retardation, brain defects (reduced anterior hippocampus, corpus callosum hypoplasia, enlarged ventricles), and behavioral abnormalities. These mice also exhibit disruptions in glutamate, dopamine, and serotonin signaling pathways, highlighting NPAS3’s essential role in brain development. At least three isoforms of NPAS3 are known.
