Target Information
Pyridoxal kinase (PDXK) converts vitamin B6 to pyridoxal-5-phosphate (PLP), an essential cofactor in amino acid and neurotransmitter metabolism. The PDXK gene encodes a 312-amino acid polypeptide with pyridoxal kinase activity. A major 1.5-kb PDXK transcript is expressed in all tissues tested and shows circadian oscillations. In mouse liver and brain, Pdxk expression is regulated by the PAR bZIP transcription factors (Dbp, Hlf, Tef), which also oscillate in expression. Mice lacking all three transcription factors exhibit reduced levels of brain PLP, serotonin, and dopamine, and are prone to spontaneous and audiogenic epilepsies.
