Product Specific Information

Description:
WM-53 monoclonal antibody reacts with human CD33 (GP67, P67), a 67 kDa type I transmembrane glycoprotein of the Siglec family. CD33 is expressed in the hematopoietic compartment, including monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human lysed whole blood. Recommended usage: 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, incorporating novel nucleic acid dye structures for engineered fluorescence with minimized spillover and spread.

Target Information

CD33 is a transmembrane protein of the Siglec family containing ITIM motifs that recruit SHP-1 and SHP-2 phosphatases. It is expressed on myelomonocytic lineage cells, binds sialic acid residues on N- and O-glycans, and is a therapeutic target for acute myeloid leukemia. CD33 is found on granulocyte and macrophage precursors but not on pluripotent stem cells, making it a useful marker for distinguishing myelogenous leukemia cells. Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
