Product Specific Information

The WM-53 monoclonal antibody reacts with human CD33, also known as GP67 and P67, a 67 kDa type I transmembrane glycoprotein belonging to the Siglec (sialic acid-binding Ig superfamily lectin) family. It is expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested for flow cytometric analysis of normal human lysed whole blood. Recommended use: 5 µL (0.8 µg) per test in a final volume of 100 µL. Cell number: 10⁵–10⁸ cells/test.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) enhances staining performance with NovaFluor conjugates.

• Add 5 µL per sample (regardless of number of NovaFluors).
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing nucleic acid dye structures engineered for optimized spectral signatures with minimal spillover and spread.

Target Information

CD33 is a transmembrane protein of the Siglec family containing immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases. It is expressed on myelomonocytic lineage cells, binds sialic acid residues on cell surfaces, and serves as a therapeutic target for acute myeloid leukemia.
Found on granulocyte and macrophage precursors in bone marrow but not on pluripotent stem cells. CD33 is also expressed on peripheral monocytes and is useful for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
