Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 up-regulates stress response genes such as p21waf1, leading to G1 or G2/M arrest.

The structure of p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is important for p53 activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells and functions as an anticancer factor. It is also linked to aging, oxidative stress, and cognitive processes such as long-term potentiation (LTP).

Mutations in the TP53 gene are common in human cancers and can lead to loss of tumor suppressor activity. Germline mutations are also associated with cancer-prone syndromes such as Li-Fraumeni syndrome.
