Target Information

The Wiskott-Aldrich syndrome family of proteins share similar domain structures and are involved in signal transduction from cell surface receptors to the actin cytoskeleton. These proteins interact with multiple partners and are regulated by various stimuli. They associate with the small GTPase Cdc42 and the Arp2/3 complex, both critical for actin filament formation and cytoskeletal organization.

Wiskott-Aldrich syndrome is a rare, X-linked recessive disorder characterized by immune dysregulation and microthrombocytopenia, caused by mutations in the WAS gene. The WAS gene product is a cytoplasmic protein expressed exclusively in hematopoietic cells, which exhibit signaling and cytoskeletal abnormalities in affected patients. An alternative transcript variant with a different 5′ UTR sequence has been reported, though its full-length structure remains unclear.
