Target Information

The LRP2 (low density lipoprotein-related protein 2), also known as megalin, is a multi-ligand endocytic receptor expressed mainly in absorptive epithelial tissues such as the kidney.
This glycoprotein includes:

• A large amino-terminal extracellular domain
• A single transmembrane domain
• A short cytoplasmic tail

It binds various macromolecules including albumin, apolipoproteins B/E, and lipoprotein lipase.
LRP2 plays a critical role in the reuptake of ligands such as lipoproteins, sterols, vitamin-binding proteins, and hormones.
It also participates in cell-signaling, interacting with extracellular ligands (e.g., parathyroid hormones, sonic hedgehog) and intracellular partners (e.g., MAP kinase scaffold proteins, JNK interacting proteins).
Recycling of LRP2 is regulated by phosphorylation of its cytoplasmic domain.
Mutations in this gene are associated with Donnai-Barrow syndrome (DBS) and facio-oculoacoustico-renal syndrome (FOAR).
