Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly during late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that allows binding to euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.
Ki-67 shows cell cycle–dependent localization: perinucleolar in G1, nuclear matrix in G2, and chromosomal during M phase.
Because Ki-67 is absent in G0 cells and increases from G1 through mitosis, it serves as a reliable proliferation marker. Ki-67 antibodies help determine the growth fraction in neoplasms, correlating strongly with histological tumor grade. It is also widely used as a neuronal marker of cell cycling and proliferation.
