Product Specific Information

  • For immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases. It associates with euchromatin, centromeric heterochromatin, and the nucleolus through its forkhead-associated (FHA) domain.
Ki-67 shows cell cycle-dependent localization: perinucleolar in G1, nuclear matrix in G2, and chromosomal during M phase. It is widely used as a proliferation marker since it is absent in G0 cells and increases from G1 through mitosis.
In neoplastic tissues, Ki-67 index correlates strongly with tumor grade and is comparable to the thymidine-labelling index. It is routinely used as a neuronal marker for cell cycling and proliferation.
