  • CD4 RPE reagent: IgG2a (rat)
  • Clone CA17.2A12 recognizes canine CD3 epsilon, expressed on mature T lymphocytes.
  • Clone YKIX302.9 recognizes canine CD4, expressed by lymphocyte subsets and canine neutrophils.
  • For FACS analysis: use 10 µL of the suggested working dilution to label 1×10⁶ cells in 100 µL.
  • Reconstitute with 1 mL distilled water. After reconstitution, store product undiluted at 4°C in the dark.

Target Information

The CD3 complex is crucial in transducing antigen-recognition signals into the cytoplasm of T cells and in regulating the cell surface expression of the TCR complex. T cell activation through the antigen receptor (TCR) involves the cytoplasmic tails of the CD3 subunits (γ, δ, ε, ζ). These subunits are structurally related members of the immunoglobulin superfamily encoded by closely linked genes on human chromosome 11.
CD3 subunits have long cytoplasmic tails that associate with signal transduction molecules via a double tyrosine-based motif. CD3 may play roles in TCR-induced growth arrest, cell survival, and proliferation. The CD3 antigen is found on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Decreased T lymphocyte percentages may occur in autoimmune diseases.

CD4 antigen is involved in recognition of MHC class II antigens and serves as a receptor for HIV. It is present on most T helper cells and normal thymocytes, associated with p56lck tyrosine kinase. CD4 expression is common in lymph nodes and tonsils.

CD44 is a type I transmembrane glycoprotein, also known as Phagocytic Glycoprotein-1 (pgp-1) and HCAM, functioning as a receptor for hyaluronate. It exists in multiple isoforms due to alternative RNA splicing and is involved in leukocyte adhesion to endothelial and stromal cells and extracellular matrix.
