Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation.
p53 up-regulates specific genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, it includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation of p53 at various sites, including Ser15 and Ser392, plays critical roles in activation, stabilization, and tumor formation. p53 is expressed at low levels in normal cells but highly in transformed cells, contributing to malignancy.
Mutations in TP53 lead to loss of tumor suppressor activity and are associated with multiple human cancers, including Li-Fraumeni syndrome.
