Target Information
BCL-2 is a key regulator of apoptosis that functions to either inhibit or promote cell death. Members of the BCL-2 family can form dimers to modulate apoptosis. Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect. Bax and Bak are critical for cytochrome c release from mitochondria, initiating apoptosis. Bax exerts a pro-apoptotic effect, while constitutive expression of BCL2, such as translocation to the Ig heavy chain locus, is associated with follicular lymphoma. In follicular lymphoma, neoplastic germinal centers express high levels of BCL-2 alpha protein, whereas normal centers are negative. Two transcript variants of BCL-2 differ in their C-terminal ends. Overexpression of BCL-2 has been linked to cancers such as B-cell lymphoma and prostate cancer.
