Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor in the TNFR superfamily. It also exists in a soluble form (~26 kDa). Upon stimulation, FAS aggregates its intracellular FAS-associated death domains (FADD), forming the death-inducing signaling complex (DISC) and activating caspases, leading to apoptosis.

FAS can activate NF-kappaB, MAPK3/ERK1, MAPK8/JNK, and is involved in proliferative signaling in fibroblasts and T cells. Multiple alternatively spliced isoforms exist, some lacking the transmembrane domain, which may negatively regulate apoptosis. The Fas/Fas ligand system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a role in antiviral immune responses.
