Product Specific Information
Specific for the ~160 kDa MerTK protein phosphorylated at Tyr749/753/754. The immunolabeling is completely eliminated by treatment with lambda-phosphatase.
Due to post-translational modifications of the Mer protein, a significant shift in molecular weight is seen from the predicted molecular weight of 110 kDa.
For optimal results, immunoprecipitation is recommended due to the 91% homology of the related receptor tyrosine kinase, Axl, that runs at ~140 kDa.
Target Information
MERTK (cMER) is a tyrosine kinase proto-oncogene involved in the retinal pigment epithelium (RPE) phagocytosis pathway, which is implicated in human retinal disease.
It regulates many physiological processes including cell survival, migration, differentiation, and phagocytosis of apoptotic cells (efferocytosis).
Ligand binding at the cell surface induces autophosphorylation of MERTK on its intracellular domain, providing docking sites for downstream signaling molecules.
Following activation by ligand, MERTK interacts with GRB2 or PLCG2 and induces phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1.
MERTK signaling plays a role in macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization, and engulfment.
In the retinal pigment epithelium (RPE), MERTK functions as a regulator of rod outer segment fragment phagocytosis.
It also inhibits Toll-like receptor (TLR)-mediated innate immune responses by activating STAT1, which induces production of SOCS1 and SOCS3.
