Target Information
TPA converts the abundant, but inactive, zymogen plasminogen to plasmin by hydrolyzing a single Arg-Val bond in plasminogen. By controlling plasmin-mediated proteolysis, it plays an important role in tissue remodeling and degradation, cell migration, and other physiological and pathological events.
TPA specifically cleaves the Arg-Val bond in plasminogen to form plasmin, and is comprised of a heterodimer of chain A and chain B held by a disulfide bond.
It binds to fibrin with high affinity, increasing catalytic efficiency 100–1000 fold due to enhanced affinity for plasminogen. Binding to heparin, laminin, and fibronectin also enhances activation.
TPA interacts with mannose receptor and LRP1, which are involved in clearance, and binds to annexin II and cytokeratin 8.
Interactions on endothelial and vascular smooth muscle cells (VSMC) stimulate plasminogen activation 100-fold and reduce inhibition by PAI-1 by 30-fold.
