Target Information
Specialized cells at the midline, which separates the left and right halves of the CNS, play key roles in directing growth cone behavior. In vertebrates, insects, and C. elegans, midline cells produce guidance cues such as nectins and slit, functioning as attractants and repellents. Robo (Roundabout) proteins act as receptors for Slit and are conserved from fruit flies to mammals. Robo1 and Robo2 are human homologs of Drosophila Robo; Robo1 is also homologous to C. elegans sax3, and Robo2 to the zebrafish gene astray. Robo functions as a gatekeeper, regulating axon crossing and responsiveness to guidance cues.
