Target Information
14-3-3는 산성이고 수용성인 단백질 계열로, 효모부터 포유류까지 아미노산 서열이 고도로 보존되어 있으며 모든 진핵세포에서 발현됩니다. 포유류에서는 beta, gamma, epsilon, eta, zeta, sigma, tau/theta의 7가지 아이소폼이 확인되며, 각각 독립적인 유전자에 의해 발현되어 homo- 및 hetero-dimer 형태의 컵 모양 구조를 형성합니다.
14-3-3 단백질은 100개 이상의 결합 파트너와 상호작용하며, 신호전달, 세포주기, 전사 조절, 세포 증식, 세포사멸, 이온 채널 생리 등 다양한 생물학적 과정에 관여합니다. 대부분의 14-3-3 결합은 표적 서열 내 serine 또는 threonine 잔기의 인산화가 필요합니다. 이 단백질은 뇌에서 풍부하게 발현되며, 다양한 신경학적 질환 환자의 뇌척수액에서도 검출된 바 있습니다.
