Target Information
cAMP is a signaling molecule important for a variety of cellular functions. It activates the cAMP-dependent protein kinase, which transduces signals through phosphorylation of different target proteins. The inactive kinase holoenzyme is a tetramer composed of two regulatory and two catalytic subunits. Upon cAMP binding, the holoenzyme dissociates into a regulatory dimer bound to four cAMP molecules and two free catalytic subunits.
Four different regulatory and three catalytic subunits have been identified in humans. The PRKAR1A gene encodes one of the regulatory subunits. This protein acts as a tissue-specific extinguisher, down-regulating expression of seven liver genes in hepatoma × fibroblast hybrids. Mutations in PRKAR1A cause Carney complex (CNC). The gene can fuse to the RET proto-oncogene to form the thyroid tumor-specific chimeric oncogene PTC2. A nonconventional nuclear localization sequence (NLS) suggests a role in DNA replication via nuclear transport of the second subunit of Replication Factor C (RFC40). Three alternatively spliced transcript variants encoding the same protein have been observed.
