Target Information

cAMP is a signaling molecule important for a variety of cellular functions. It activates cAMP-dependent protein kinase, which transduces signals through phosphorylation of target proteins. The inactive kinase holoenzyme is a tetramer composed of two regulatory and two catalytic subunits. cAMP causes dissociation of the holoenzyme into regulatory dimers and free catalytic monomers.

Four regulatory and three catalytic subunits have been identified in humans. The PRKAR1A gene encodes one of the regulatory subunits, acting as a tissue-specific extinguisher that down-regulates liver gene expression in hepatoma × fibroblast hybrids. Mutations in this gene cause Carney complex (CNC). It can fuse with the RET protooncogene to form the thyroid tumor-specific chimeric oncogene PTC2. A nonconventional nuclear localization sequence (NLS) suggests a role in DNA replication via nuclear transport of RFC40. Three alternatively spliced transcript variants encoding the same protein have been observed.
