Target Information

Calcium-permeable channels such as TRPV6 are involved in neurotransmission, muscle contraction, and exocytosis by mediating intracellular calcium signaling. Depending on tissue type, calcium transport may be regulated by vitamin D, parathyroid hormone (PTH), or calcitonin (CALCA).
TRPV6 acts as a calcium-selective cation channel involved in Ca²⁺ uptake in various tissues, including intestinal reabsorption. The channel is activated by low internal calcium levels and exhibits inward rectification.
Inactivation occurs via rapid Ca²⁺-dependent and slower Ca²⁺-calmodulin-dependent mechanisms, possibly regulated by phosphorylation. In vitro, it is slowly inhibited by Mg²⁺ in a voltage-independent manner.
TRPV6 forms heteromeric assemblies with TRPV5, altering channel properties. TRPV5–TRPV6 heteromultimers exhibit voltage-dependent gating. TRPV6 interacts with TRPV5, S100A10, ANAX2–S100A10 heterotetramer, BSPRY, and calmodulin. Interaction with S100A10 is essential for plasma membrane trafficking.
