Target Information
GRP78 is a 78 kDa glucose-regulated protein belonging to the heat shock protein family (HSP70). It resides in the endoplasmic reticulum (ER) and transiently associates with newly synthesized secretory and membrane proteins. GRP78 interacts with mutant and misfolded proteins, marking them for degradation.
The conserved sequence Lys-Asp-Glu-Leu (KDEL) at the C-terminus of GRP78 and other ER proteins (GRP94, PDI) ensures their recycling back to the ER via the KDEL receptor.
GRP78 plays a key role in protein folding and degradation of misfolded proteins through its interaction with DNAJC10, maintaining cellular homeostasis and preventing apoptosis.
GRP78 levels serve as reliable biomarkers of hypoglycemia and have neuroprotective functions under oxidative stress. Reduced expression is observed in Alzheimer’s disease and associated with PS1 gene mutations. Induction of GRP78 is linked to drug resistance in cancer therapy.
