Target Information
The expression pattern of the Prox1 homeobox gene suggests roles in various embryonic tissues, including the lens. Prox1 mRNA is present in many human tissues, with the highest level in the lens. Homozygous Prox1−/− mice die midgestation due to multiple developmental defects, including lens abnormalities. Prox1 inactivation leads to aberrant cellular proliferation, downregulation of Cdkn1b and Cdkn1c, misexpression of E-cadherin, and increased apoptosis, resulting in defective lens formation.
Prox1 is expressed in endothelial cells that form the lymphatic system, acting as a specific regulator of lymphatic development. It also regulates hepatocyte migration in mice; loss of Prox1 causes smaller livers with fewer clustered hepatocytes. In the embryonic retina, Prox1 controls progenitor cell cycle exit and differentiation, playing a key role in proliferation and fate determination.
