Target Information
Gamma-aminobutyric acid (GABA) is the primary inhibitory neurotransmitter in the central nervous system, causing hyperpolarization of the membrane through the opening of a Cl⁻ channel associated with the GABA-A Receptor (GABA-A R) subtype.
GABA-A Rs are important therapeutic targets for sedative, anxiolytic, and hypnotic agents, and are implicated in diseases such as epilepsy, anxiety, depression, and substance abuse.
The GABA-A R is a multimeric subunit complex consisting of six alphas, four betas, and four gammas, plus alternative splicing variants. Expression of alpha and beta subunits results in functional GABA-A Rs sensitive to GABA, while coexpression of a gamma-subunit is required for benzodiazepine modulation.
Phosphorylation of beta subunits modulates GABA-A R function, and different alpha subunits may mediate distinct benzodiazepine effects in the brain.
