Target Information
CD11b (integrin alpha-M, ITGAM, integrin alpha-X, ITGAX) is a 165 kDa adhesion molecule that associates non-covalently with integrin beta-2 (CD18). The CD11b/CD18 heterodimeric complex, also known as integrin alpha-M beta-2, Mac-1, and CR3 (complement receptor 3), is expressed on the surface of monocytes/macrophages, granulocytes, activated lymphocytes, subsets of NK cells, dendritic cells, and microglia.
CD11b/CD18 functions as a receptor for multiple ligands including ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X. These interactions are crucial for cell-cell and cell-matrix adhesion.
CD11b expression varies among tissues: 8% in spleen cells, 44% in bone marrow cells, and <1% in thymocytes. It is a common microglial marker, with expression increasing during monocyte maturation. Peritoneal macrophages express higher CD11b levels than splenic macrophages.
CD11b dysfunction is associated with systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune diseases.
