ACAP1 is a transport carrier protein containing three ANK repeats, an Arf-GAP domain, a BAR domain, and a PH domain that binds phospholipids. It acts as a GTPase-activating protein (GAP) for ADP ribosylation factor 6 (ARF6), essential for clathrin-dependent export of proteins from recycling endosomes to the trans-Golgi network and cell surface.
It mediates binding to phosphatidylinositol 3,5-bisphosphate (PIP2) or phosphatidylinositol 3,4,5-trisphosphate (PIP3) containing membranes.
ACAP1 is recruited along with ACAP2 to PDGF-induced dorsal membrane ruffles in NIH 3T3 mouse fibroblasts, and its overexpression inhibits ruffle formation.
It is widely expressed in most tissues, with high expression in lung and spleen, and low expression in heart, kidney, liver, and pancreas.
