Product Specific Information
PA1-1059 detects IRS-1 in mouse and human cells.
It has been successfully used in Western blot procedures, detecting a ~170 kDa protein representing IRS-1 from 3T3 L1 cell extract.
The PA1-1059 immunogen is a synthetic peptide corresponding to residues D(631) Y M P M S P K S V S A P Q Q I(646) of human IRS-1, with 100% conservation in rats and mice.
This peptide (Cat. # PEP-193) is available for use in neutralization and control experiments.
Target Information
IRS-1, a major substrate of the insulin receptor, is phosphorylated in response to stimulation by insulin, IGF-1, and IL-4.
Phosphorylation occurs on serine, threonine, and tyrosine residues across various tissues.
Casein kinase II mediates a portion of insulin-stimulated serine/threonine phosphorylation of IRS-1 in vivo, with Thr 502 and Ser 99 identified as major phosphorylation sites.
IRS-1 contains three putative 14-3-3 binding sites (Ser 270, Ser 374, Ser 641), where the motif around Ser 270 is located in the phosphotyrosine binding domain responsible for interaction with the insulin receptor.
