배경 (Background)
Defects in GATA1 are associated with:
- X-linked dyserythropoietic anemia and thrombocytopenia (XDAT) [MIM:300367]: characterized by abnormal erythrocyte morphology and platelet deficiency.
- X-linked thrombocytopenia with beta-thalassemia (XLTT) [MIM:314050]: features thrombocytopenia, platelet dysfunction, hemolysis, and imbalanced globin synthesis.
Functional Domains:
- Contains two GATA-type zinc fingers that cooperate for specific and stable DNA binding.
- The first finger contributes to specificity and stability; the second is essential for DNA binding.
Function:
- Acts as a transcriptional activator and general switch factor for erythroid development.
- Binds to DNA consensus sequence [AT]GATA[AG] in regulatory regions of globin and other erythroid genes.
Post-translational Modifications (PTM):
- Highly phosphorylated on serine residues.
- Phosphorylation on Ser-310 increases during erythroid differentiation.
- Phosphorylation on Ser-142 promotes sumoylation on Lys-137.
- Sumoylation by SUMO1 does not affect transcriptional activity.
Protein Interactions:
- Interacts with ZFPM1, GFI1B, and PIAS4 (enhances sumoylation and represses transactivation activity).
- Tissue specificity: Erythrocytes.
