Background
- Cofactor: Binds 2 zinc ions and 4 calcium ions per subunit.
- Disease Association: Defects in MMP13 cause spondyloepimetaphyseal dysplasia type 2 (SEMD2) [MIM:602111], also known as spondyloepimetaphyseal dysplasia type Missouri. SEMDs are skeletal disorders characterized by defective growth and modeling of spine and long bones, involving both epiphyses and metaphyses.
- Domain: The conserved cysteine in the cysteine-switch motif binds catalytic zinc, inhibiting the enzyme. Activation occurs upon dissociation of cysteine from zinc after activation-peptide release.
- Function: Degrades collagen type I; does not act on gelatin or casein; may play a role in tumor processes.
- Similarity: Belongs to peptidase M10A family; contains four hemopexin-like domains.
- Tissue Specificity: Seems specific to breast carcinomas.
