Background

Dipeptidyl-peptidase 1은 thiol protease로서 다양한 dipeptide 기질에 대해 활성을 가지며, serine protease(예: elastase, cathepsin G, granzymes A/B)를 활성화합니다. 결함은 Haim-Munk syndrome (HMS), juvenile periodontitis (JPD), Papillon-Lefevre syndrome (PLS) 등과 관련이 있습니다.
해당 단백질은 리소좀에 위치하며, 폐, 신장, 태반에서 높은 발현을 보이고, 대장, 소장, 비장, 췌장에서 중간 수준으로 발현됩니다.

Enzyme regulation:

• cysteine peptidase inhibitors (mersalyl acid, iodoacetic acid, cystatin)에 의해 강하게 억제됨
• N-ethylmaleimide, Gly-Phe-diazomethane, TLCK, TPCK, dithiodipyridine(low pH)에도 억제됨
• PMSF, bestatin, 금속 이온 킬레이터에는 억제되지 않음

PTM: N-glycosylated; exclusion domain은 position 58 또는 61에서 절단됨
