Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are naturally low, representing a small fraction of total immunoglobulins. IgE plays a role in allergic responses by binding to mast cells and basophils via Fc epsilon receptor I, leading to the release of histamine and other mediators. IgE is produced by terminally differentiated plasma B cells and basophils, existing both as soluble and membrane-bound forms. IgE21 recognizes both forms.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested for flow cytometry on normal human peripheral blood cells. Use 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Super Bright 600 is a tandem dye excited by the 405 nm violet laser and emits at 600 nm. Recommended detection filter: 610/20 bandpass. Ensure the instrument supports this fluorochrome.

When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

IgE is one of the five classes of mammalian immunoglobulins. It consists of two heavy (epsilon) chains with four constant domains and two light chains. IgE is essential in type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding to Fc receptors on basophils and mast cells. Crosslinking of receptor-bound IgE triggers degranulation, releasing mediators that cause allergic symptoms. IgE also contributes to immunity against parasites.
