Target Information
Raf-1 (c-Raf) is a MAP kinase kinase kinase (MAP3K) that functions downstream of the Ras family of membrane-associated GTPases. Once activated, Raf-1 phosphorylates and activates MEK1 and MEK2, which subsequently activate ERK1 and ERK2. Activated ERKs regulate gene expression controlling cell cycle, apoptosis, differentiation, and migration. Mutations in RAF1 are associated with Noonan syndrome 5 and LEOPARD syndrome 2.
