Carrier-Free
Reconstitution:
Centrifuge the vial briefly before opening. Reconstitute the lyophilized protein in sterile 20 mM Citrate Buffer (pH 5.0) or 10 mM Acetic Acid to a concentration of 0.1–0.5 mg/mL to regain full activity. Apportion the reconstituted protein into working aliquots and store at ≤ -20°C. Make further dilutions in medium or buffered solution containing carrier protein (e.g., PBS with 0.1% BSA).
Storage:
Store the lyophilized protein at 2–8°C, preferably desiccated. After reconstitution, store aliquots at ≤ -20°C (not in a frost-free freezer). Avoid repeated freeze-thaw cycles.
Target Information
Sonic Hedgehog (SHH)는 배아 발생 동안만 발현되며, 초기 배아의 패턴 형성에 중요한 역할을 합니다. 특히 신경관의 복측부, 사지의 전후축, 그리고 복측 체절의 패턴 형성에 관여하는 주요 유도 신호로 알려져 있습니다.
이 단백질은 전구체 형태로 생성되어 자가 촉매적으로 절단되며, N-말단은 신호 기능을, C-말단은 전구체 처리에 관여합니다. 또한 C-말단 산물은 N-말단 산물에 콜레스테롤기를 공유 결합시켜 세포 표면에 부착시킵니다.
이 단백질 또는 그 신호 경로의 결함은 전뇌가 좌우로 분리되지 않는 선천성 질환인 holoprosencephaly (HPE)를 유발할 수 있으며, VACTERL 증후군과도 관련이 있을 수 있습니다.
