Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon stress, p53 activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.
Structurally, p53 consists of an N-terminal transactivation domain, a central DNA-binding domain, an oligomerisation domain, and a C-terminal regulatory domain. Phosphorylation at Ser15 is critical for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation.
p53 plays a key role in blocking proliferation of damaged cells and acts as an anticancer agent. It is also linked to aging, oxidative stress, and learning deficits via effects on LTP (Long Term Potentiation).
Normal cells express low levels of p53, whereas transformed cells show high expression, contributing to malignancy. Mutations in TP53 lead to loss of DNA-binding and tumor suppressor activity, occurring both as somatic and germline mutations (e.g., Li-Fraumeni syndrome).
