Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimer or CD8αα homodimer. CD8a is found on thymocytes, mature T cells, and NK cells, binding to MHC class I and associating with p56lck to mediate T-cell development and activation.

Preliminary testing shows OKT8, RPA-T8, and HIT8a clones do not compete for epitopes on human leukocytes, indicating distinct binding sites.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells (5 µL (0.1 µg)/test).
A test is defined as the amount of antibody that stains cells in 100 µL final volume (10⁵–10⁸ cells/test).

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

Buffer Recommendation:
CellBlox Plus Blocking Buffer (Product # C001T06F01) is required for optimal staining.
Add 5 µL per sample before combining with cells or antibody cocktails.

CD8 (Cluster of Differentiation 8)
CD8 is a type I transmembrane glycoprotein of the immunoglobulin receptor family, essential in T-cell differentiation and activation.
It functions as a co-receptor with TCR for MHC class I molecules. CD8α chain binds MHC-I and is expressed on T cells, NK cells, monocytes, and dendritic cells.
Upon MHC-I/peptide ligation, CD8 recruits Lck kinase, triggering cytokine production and CTL activation.
Activated CTLs play a key role in pathogen and tumor cell clearance. Differentiation is enhanced by IL-2, IL-12, and TGF-β1.
