Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule (~32–34 kDa), expressed as a heterodimer (CD8αβ) or homodimer (CD8αα). CD8a is found on most thymocytes and a subset of mature T cells and NK cells. It binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.

Preliminary testing shows OKT8 does not compete with other anti-human CD8 clones (RPA-T8, HIT8a) for binding, indicating distinct epitopes.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (0.1 µg)/test in 100 µL final volume. Optimal cell count: 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
  • CellBlox Plus Blocking Buffer (Product # C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Based on Phiton™ technology, utilizing engineered nucleic acid dye structures for optimized fluorescence with minimal spillover and spread.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin family. It functions as a co-receptor with TCR for MHC class I molecules. CD8 is mainly expressed as a disulfide-linked heterodimer (CD8αβ) on T cells, and as a homodimer (CD8αα) on subsets of T cells, NK cells, monocytes, and dendritic cells.

Upon MHC-I/peptide ligation, CD8 recruits Lck kinase, leading to lymphokine production, motility, and CTL activation. Activated CTLs are essential for pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
