Target Information
WASP (Wiskott-Aldrich syndrome protein) and N-WASP are downstream effectors of Cdc42 involved in actin polymerization and cytoskeletal organization. The WASP family includes VASP and Mena, which localize at focal adhesions and regulate actin cytoskeleton dynamics. WAVE proteins, related to WASP family members, mediate actin reorganization downstream of Rho family GTPases.
WAVE1 and WAVE2 regulate membrane ruffling by inducing actin filament clusters in response to Rac activation. WAVE proteins promote actin polymerization through cooperation with the Arp2/3 complex. WAVE1 (also known as SCAR) is primarily expressed in the brain, while WAVE2 is broadly expressed, especially in peripheral blood leukocytes. WAVE3 forms a multiprotein complex linking receptor kinases and actin, mediating signals that influence cell shape, motility, and function.
