Target Information
TNK2 (ACK), also known as activated Cdc42-associated kinase, is a non-receptor tyrosine kinase that signals downstream of Cdc42. It plays a key role in tumorigenesis and metastasis, particularly in prostate cancer. TNK2 mediates CDC42-dependent cell migration via phosphorylation of BCAR1 and is involved in synaptic function and brain development. It activates AKT1 by phosphorylating Tyr-176, and phosphorylates AR (Tyr-267, Tyr-363), WWOX (Tyr-287), and MCF2, enhancing its GEF activity toward Rho family proteins. TNK2 regulates AXL receptor levels, confers metastatic properties, and promotes tumor growth by modulating tumor suppressors and survival factors.
