Product Description
이 키트는 Illumina 플랫폼에서 차세대 시퀀싱(NGS)에 사용되는 16S V4-V5 앰플리콘 라이브러리 준비를 위한 모든 분자 시약(프라이머, 효소 혼합물, 인덱스, 버퍼)을 제공합니다.
AMPure XP Magnetic Beads(고객 제공)를 이용한 PCR 정제 단계의 지침이 포함되어 있으며, 다양한 DNA 시료(대변, 토양, 물, 타액, 식물, 소변, 피부, 구강, 비강 등)에 적용 가능합니다.
라이브러리 준비 절차는 약 4시간 내 완료됩니다.
DNA는 먼저 16S rRNA 유전자의 V4-V5 영역을 증폭하기 위한 타깃 PCR을 거치며, 이후 AMPure XP beads를 사용해 정제합니다. 듀얼 인덱스 프라이머를 제한 사이클 PCR에 사용하고, 5’ 및 3’ 바코드 어댑터가 부착된 앰플리콘을 최종 정제 후 정량, 풀링, 시퀀싱에 사용합니다.
