Target Information
Sterol regulatory element-binding proteins (SREBPs) are transcription factors belonging to the basic helix-loop-helix leucine zipper family of DNA-binding proteins. Three isoforms are identified in mammalian tissues with distinct structures and functions.
SREBP-1a and SREBP-1c are products of alternative promoter usage of the SREBP-1 gene, while SREBP-2 is transcribed from a separate gene.
SREBPs exist as 120 kDa inactive precursors in the ER membrane and, upon activation, are cleaved in the Golgi to form 60–78 kDa active fragments.
In liver and adipose tissues, SREBPs regulate lipid and cholesterol synthesis by controlling the transcription of related genes. SREBP-1 mainly regulates triglyceride synthesis, whereas SREBP-2 is involved in cholesterol metabolism.
